ORALLY DISINTEGRATING TABLETS - ADVATAB® AND MICROCAPS™

Eurand's AdvaTab is a new generation of fast dissolving tablet that offer distinct advantages and unique applications. Advatab enables the development of high - dose products, therapeutic applications, controlled release formulations, efficient manufacturing and flexible packaging options.

AdvaTab is supported by our Microcaps technology platform (coacervation microencapsulation) that taste-masks drug particles by polymer encapsulation to create particles (typically 150 microns to 300 microns in size) that are suitable for use in orally disintegrating tablets. In addition, Eurand has developed proprietary and innovative coating techniques to enable the development of taste-masked products without compromising bioequivalence to a conventional tablet or capsule dosage form.

AdvaTab tablets disintegrate quickly producing a smooth, creamy mixture of drug granules and carrier, which is easily swallowed with a sweet, pleasant taste. AdvaTab tablets can carry high doses of drug; for example, Eurand has developed a 500 mg acetaminophen product.

CUSTOMIZED RELEASE TECHNOLOGY

Eurand's customized release technology platform can be applied to a wide range of molecules and has a proven track record throughout the industry. Eurand works to achieve the desired pharmacokinetic profiles for a specific compound, improving the therapeutic value of the product. Eurand utilizes three multiparticulate technologies and one solid, monolithic tablet controlled release product. Multiparticulate formulations offer additional advantages:

• Flexible dose adjustment for clinical studies
• Less patient variability due to gastric emptying times
• Simple creation of multiple product strengths
• Combinations of drugs

Eurand's Diffucap technology provides effective drug release by layering active drugs onto a neutral core such as sugar spheres, crystals or granules followed by one or more rate-controlling, functional membranes. Eurand can customize any combination of sustained, pulsatile and immediate release profiles depending on the client's specific requirements for the product.

At 1.5mm to 2.0mm in diameter, Eurand's Minitabs tablets can contain gel-forming excipients that control drug release rate or can be a reservoir of drug formulation. Additional membranes can be added to further enhance the control release rate. Minitabs tablets are encapsulated into a final dosage form to achieve high drug loads.

Orbexa technology produces beads of a controlled size and density using granulation extrusion and spheronization techniques. The resultant beads can be coated with release rate-controlling membranes for additional release rate control and may be filled into capsules or provided in sachet form.

Applicable to both soluble and insoluble products, Diffutab technology incorporates a blend of hydrophilic polymers that controls drug release through diffusion and erosion of a matrix tablet.

ENHANCED SOLUBILITY TECHNOLOGY - BIORISE

Eurand's proprietary Biorise™ technology optimizes the delivery of drugs that are poorly absorbed due to their inherent solubility problems. By optimizing the therapeutic performance of these types of drugs with Biorise, Eurand offers numerous opportunities to its partners to improve their product portfolio, rescue new chemical entities and provide better medicines to their customers.

The basis for Biorise is that the solubility of nanocrystalline and amorphous forms of compounds are higher than those of unmodified or even micronized forms. Therefore, absorption rate and absolute bioavailability of the drug can be increased if drug permeability is sufficient. A poorly soluble, yet permeable crystalline drug is considered activated when it undergoes a change in its physical state that improves its biopharmaceutical properties. Eurand's iorise technology is designed to achieve this change by applying either a solid or liquid state activation process.
