Gentamicin Determination with a Metrohm Ion Chromatography System According to Ph. Eur.

Gentamicin is a broad spectrum water-soluble aminoglycoside antibiotic, which is produced by fermentation of Micromonospora purpurea. It is a complex mixture of four major components: Gentamicin C1, C1a, C2 and C2a. In addition the minor component C2b is also of interest, although its concentration is very low.

An HPLC determination requires a very complex system with quaternary gradient and derivatization of the sample prior to injection. This is very time and cost consuming. Metrohm offers therefore a determination with a pulsed amperometric detector and isocratic elution on a polystyrene column. The sample is injected directly into the chromatography system. Long time derivatization is no more necessary. If the sample has a very high loaded matrix, it is no problem in a Metrohm system to implement MISP (Metrohm Inline Sample Preparation technique) such as Inline Filtration or Inline Dialysis. The method is according to the European Pharmacopoeia (Ph. Eur.).

The «MIC-9 Advanced» IC system combines the two most important electrochemical detection methods: conductivity measurement and amperometric determination. Conductometry with the 819 High-Performance Detector analyzes either cations with electronic suppression or anions with chemical suppression down to the lower ppb range. The pulsed amperometric detector of the 817 Bioscan determines readily oxidizable or reducible substances. The detection sensitivities are equal to those of the conductivity detector.

With the «MIC-9 Advanced» you will receive a completely flexible system, with which you can work with two channels, either in parallel or independently. The only common system component is the 838 Advanced IC Ultrafiltration Sample Processor which, thanks to its ultrafiltration cell, is outstandingly suitable for sample preparation.
