Vivapure AdenoPACK 20 - Small-scale Virus Purification

Sartorius AG has expanded its adenovirus purification range by launching a new kit for small-scale purification: Vivapure AdenoPACK 20.

With Vivapure AdenoPACK 20, Sartorius offers a kit that combines quick and easy handling with the option for processing 6 x 20ml cell culture samples in parallel. This new product not only addresses researchers working simultaneously with multiple viral constructs, but also users who need only small amounts of viruses. By contrast, the traditional cesium chloride density gradient method only works effectively with cell pellets obtained from cell culture volumes larger than 100ml.

The new product is supplied with all reagents and devices necessary for clarification, purification and concentration of type 5 adenoviruses from HEK293 cells. The rapid one-hour purification protocol is based on Sartorius proprietary ion exchange membrane adsorbers in AdenoPACK 20 spin columns. These spin column units, have the unique capability of binding viral particles efficiently and re-releasing them completely.

Compared to conventional chromatography media, membrane adsorbers feature 3000nm sized pores, thus allowing viral particles unrestricted access to the charged adsorber surface. By this method, it takes just an hour to recover up to 1 x 1012 viruses from 20ml of cell culture medium for in-vitro transfection.

Unlike conventional purification procedures utilizing cesium chloride density gradients, this method is a fast, simple and non-toxic way to purify adenoviruses. Moreover, assuming the same cell culture density, higher virus titers can be harvested after purification, which obviates the need for expensive, high-maintenance purification equipment like ultracentrifuges.

The high-capacity membrane adsorbers with porous matrices feature low differential pressures, high flow rates and low non-specific binding, and are therefore excellently suited to small-scale virus purification. For processes requiring high-performance separation and large sample volumes, the AdenoPACK kits are scalable to conform to cGMP plant requirements.
