Inclisiran is under clinical development by Novartis and currently in Phase III for Acute Coronary Syndrome. According to GlobalData, Phase III drugs for Acute Coronary Syndrome have an 18% phase transition success rate (PTSR) indication benchmark for progressing into Pre-Registration. GlobalData’s report assesses how Inclisiran’s drug-specific PTSR and Likelihood of Approval (LoA) scores compare to the indication benchmarks. Buy the report here.

GlobalData tracks drug-specific phase transition and likelihood of approval scores, in addition to indication benchmarks based off 18 years of historical drug development data. Attributes of the drug, company and its clinical trials play a fundamental role in drug-specific PTSR and likelihood of approval.

Inclisiran overview

Inclisiran (Leqvio, Sybrava) is a cholesterol-lowering agent. It is formulated as solution for subcutaneous route of administration. Leqvio is indicated in adults with primary hypercholesterolemia (heterozygous familial and non-familial) or mixed dyslipidemia, as an adjunct to diet: in combination with a statin or statin with other lipid lowering therapies in patients unable to reach LDL-C goals with the maximum tolerated dose of a statin, or alone or in combination with other lipid-lowering therapies in patients who are statin-intolerant, or for whom a statin is contraindicated. Leqvio is indicated as an adjunct to diet and maximally tolerated statin therapy for the treatment of adults with heterozygous familial hypercholesterolemia or clinical atherosclerotic cardiovascular disease, who require additional lowering of low-density lipoprotein cholesterol.

Inclisiran (ALN-PCSK9sc) is under development for the treatment of heterozygous familial hypercholesterolemia, homozygous familial hypercholesterolemia, hypercholesterolemia, pediatric hyperlipidemia, coronary artery disease (CAD), acute coronary syndrome and elevated low-density lipoprotein-cholesterol (LDL-C). It is administered through subcutaneous route in the form of solution. The drug candidate is a small interfering RNA (siRNA) which acts by targeting intracellular and extracellular PCSK9. It incorporates RNA interference with LNP technology.
