The method involves passing the sample through a size exclusion chromatography matrix to separate the free target analyte from the protein-bound target analyte. The free target analyte is then eluted from the matrix using an organic solvent, and its amount is quantified. The aqueous sample can be a biological fluid such as serum, plasma, urine, cerebrospinal fluid, or saliva. The size exclusion chromatography matrix used in the method is a macroporous cellulose polymer, and the organic solvent can be methyl tert-butyl ether, diisopropyl ether, methanol, or a combination thereof.

Furthermore, the method includes additional steps such as drying the eluted free target analyte, redissolving it without derivatizing, and quantifying it using an immunoassay procedure. Alternatively, the dried eluted free target analyte can be redissolved and derivatized before quantification using mass spectrometry. The method also allows for the addition of an isotopically labeled internal standard to the organic solvent, the aqueous sample, or the eluent for accurate quantification. Mass spectrometry, tandem mass spectrometry, or triple quadrupole mass spectrometry can be used for quantification, along with High Performance Liquid Chromatography (HPLC) separation for purification. The aqueous sample, wash solution, and organic solvent can be passed through the size exclusion chromatography matrix by gravity, centrifugation, or using negative or positive pressure.
