E. Coli Clarification

In this project, the FUNDALOOP® filter was used to clarify a lysed E. Coli culture and recover plasmid DNA. The culture (OD 100) was lysed and flocculated, creating large floating flocculates and increasing the volume fourfold. Filtration was needed to remove debris while fully recovering plasmid DNA. Depth filters were inconsistent, often clogging or failing due to culture variability, with yields of 60–75% at lab scale and up to 85% in ideal production runs. Because of these issues, the FUNDALOOP® filter was tested to ensure reliable, clog-free filtration and maximize plasmid DNA yield regardless of culture variability.
