The performance of an mRNA therapeutic depends on both the RNA molecule and the delivery system. Sequence architecture affects stability and protein expression, while formulation influences cargo protection, cellular uptake, biodistribution and tolerability. mRNA design and production

Creative Biolabs supports the design of mRNA constructs for protein replacement, immunotherapy, genome editing and other research applications. Design parameters may include the coding sequence, codon usage, untranslated regions, 5′ cap structure and poly(A) tail.

Modified nucleosides can be considered when appropriate to improve RNA performance or reduce unwanted innate immune recognition. Because their effects depend on the construct and application, sequence and chemical modifications are evaluated in the context of the complete molecule rather than treated as universal requirements.

Following template preparation, mRNA can be produced by in vitro transcription and purified to remove residual template, enzymes, unincorporated nucleotides and double-stranded RNA-related impurities. Analytical testing may include identity, integrity, purity, concentration, capping efficiency and poly(A) tail assessment. Expression studies can then determine whether the mRNA produces the expected protein in a relevant cellular system.

LNP formulation development

Lipid nanoparticles are widely used to protect mRNA from degradation and facilitate intracellular delivery. A typical LNP formulation contains an ionizable lipid, a helper phospholipid, cholesterol and a PEG-lipid. The identity and ratio of these components influence particle formation, encapsulation, stability, cellular uptake and endosomal release.

Creative Biolabs develops and screens LNP formulations using project-specific lipid compositions and preparation conditions. Studies may examine lipid ratios, RNA-to-lipid ratio, mixing parameters, buffer conditions and downstream processing.

Candidate formulations can be characterised for particle size, polydispersity, encapsulation efficiency, RNA integrity, lipid composition, morphology, surface properties and storage stability. Formulation performance is assessed alongside physical characteristics so that selection is not based on particle size or encapsulation efficiency alone.

Targeted and tissue-specific delivery

For programmes requiring preferential delivery, researchers can modify LNPs through lipid composition or by adding targeting ligands. Candidate systems may be evaluated in relevant primary cells, established cell lines or other experimental models.

In vitro studies can assess cellular uptake, endosomal escape, protein expression, biological activity and cytotoxicity. Comparative screening helps identify formulations that balance delivery efficiency with acceptable cellular responses.

Preclinical evaluation

Selected mRNA–LNP candidates can progress to preclinical studies examining biodistribution, expression kinetics, pharmacological activity and tolerability. Depending on the programme, studies may also evaluate cytokine responses, tissue-specific expression, repeat-dose effects and disease-relevant efficacy endpoints.

Analytical and biological results are reviewed together when selecting candidates. A formulation that produces high expression in one model may not offer the most appropriate distribution, stability or tolerability profile for the intended application.
